What you do is a quantitative RT-PCR using a highly specific primer towards a nonconserved region (typically the spike protein) or an antibody based test again using the spike protein.
This gives high specificity for COVID-19, so there is close to 0% false positives and if the sensitivity is high, there will also be close to 0% false negatives.
